Evaluation of the exoantigen test for identification of Histoplasma species and Coccidioides immitis cultures.

Recently it was demonstrated that certain fungal pathogens could be rapidly and accurately identified on the basis of their exoantigens. Mycelial-form cultures of the Histoplasma species consistently appear to produce either H or M antigens, or both, whereas cultures of Coccidioides immitis produce tube precipitinogen, heat-labile precipitinogen, or heat-labile F antigens. Three laboratories ea...

Limitation of the AccuProbe Coccidioides immitis culture identification test: false-negative results with formaldehyde-killed cultures.

The AccuProbe Coccidioides immitis culture identification test (CI test) yielded false-negative results with formaldehyde-killed C. immitis submitted to a reference laboratory. Further evaluation with pure or mixed cultures or stored, heat-killed cultures revealed the CI test to be highly sensitive and specific for C. immitis except when the cultures were pretreated with formaldehyde.

Immunological procedure for the rapid and specific identification of Coccidioides immitis cultures.

An immunological procedure for the rapid and specific identification of Coccidioides immitis isolates has been developed. The specificity of the procedure is based on the fact that C. immitis produces antigens that are not produced by morphologically similar fungi. The procedure involved the transfer of heavy mold-form inocula to flasks that contained small volumes of brain heart infusion broth...

Comparative evaluation of a chemiluminescent DNA probe and an exoantigen test for rapid identification of Histoplasma capsulatum.

A chemiluminescent DNA probe (Accuprobe) assay developed by Gen Probe, Inc., for the rapid identification of Histoplasma capsulatum was evaluated and compared with the exoantigen test by using 162 coded cultures including Histoplasma capsulatum var. capsulatum, Histoplasma capsulatum var. duboisii, Histoplasma capsulatum var. farciminosum, Blastomyces dermatitidis, Coccidioides immitis, Paracoc...

Collagenolytic activity of Coccidioides immitis.

Coccidioides immitis appears to be unable to digest particulate collagen when cultured on collagen-containing semisolid culture media. However, all C. immitis strains solubilized collagen when the fungus was grown in liquid suspension cultures. Moreover, sterile culture filtrates were collagenolytic in collagen-buffer-agar plate assays.
